For DNA extraction, you can use conventional kit prepared for measuring DNA (e.g. CyQUANT® or PicoGreen®) from Invitrogen. To extract/lyse the cells from QGel™ matrices after cell culture, freeze the gel discs, and subsequently use proteases (e.g. Proteinase K) to dissolve the gels and lyse the cells. For RNA extraction protocols please refer to the following publications: Kraehenbuehl T.P. et al, Biomaterials, 2008 and Lee S.T. et al, Biomaterials, 2009. Collagenase digestion, or alternatively mechanical destruction, of gels in appropriate RNA extraction buffer can be used to solubilize the gel for this purpose. Click on this FAQ to find a protocol.
